广西医科大学学报JOURNALOFGUANGXIMEDICALUNIVERSITY2023Jan;40(1)高盐诱导高血压大鼠心肌间质重构及SHIP-1和IL-6的表达研究*曾清清1,高忠兰2#,陈务贤2△,黄荣杰2△(1.广西医科大学,南宁530021;2.广西医科大学第一附属医院心血管内科,南宁530021)摘要目的:探讨高盐诱导高血压大鼠心肌间质纤维化及SH2结构域的肌醇5-磷酸酶1(SHIP-1)和白介素6(IL-6)的表达。方法:将25只SD雄性大鼠随机分为对照组和模型组,模型组给予高盐饲料(含3%NaCl)喂养12周,对照组给予普通饲料喂养12周。每两周测量大鼠尾动脉收缩压,12周后称取大鼠体重和心脏重量,计算心脏重量指数(心脏重量/体重)。采用苏木精—伊红(HE)染色及Masson染色观察心肌组织病理学改变。分别采用实时荧光定量PCR(RT-qPCR)法和免疫组化法检测SHIP-1和IL-6mRNA及其蛋白表达。结果:模型组喂养12周后的收缩压和心脏重量指数均高于对照组(P<0.05)。与对照组比较,模型组大鼠心肌组织大量炎性细胞浸润,胶原纤维显著增多,胶原容积分数(CVF)高于对照组(P<0.05)。模型组心肌组织SHIP-1mRNA及蛋白表达量低于对照组,IL-6mRNA及蛋白表达量高于对照组(均P<0.05)。结论:高盐诱导高血压大鼠出现心肌间质纤维化重构,同时SHIP-1表达下调,IL-6表达上调。关键词高盐诱导高血压;SHIP-1;IL-6;心肌间质纤维化中图分类号:R544.1文献标志码:A文章编号:1005-930X(2023)01-0054-05DOI:10.16190/j.cnki.45-1211/r.2023.01.009MyocardialinterstitialremodelingandexpressionsofSHIP-1andIL-6inhigh-salt-in⁃ducedhypertensiveratsZengQingqing1,GaoZhonglan2,ChenWuxian2,HuangRongjie2.(1.GuangxiMedicalUniversity,Nanning530021,China;2.DepartmentofCardiovascularMedicine,TheFirstAffiliatedHospitalofGuangxiMedicalUni-versity,Nanning530021,China)AbstractObjective:ToinvestigatemyocardialinterstitialfibrosisandtheexpressionsofSH2domain-contain-inginositol5-phosphatase1(SHIP-1)andinterleukin-6(IL-6)inhigh-salt-inducedhypertensiverats.Methods:Twenty-fiveSDmaleratswererandomlydividedintocontrolgroupandmodelgroup.Themodelgroupandthecontrolgroupwerefedrespectivelywithhighsaltdiet(containing3%NaCl)andordinarydietfor12weeks.Therattailarterypressurewasmeasuredonceeverytwoweeks.After12weeks,therats'bodyandheartweightwerewei...
