2012年第16卷第24期实用临床医药杂志JoumalofCliniealMedicineinPractice·11·麦冬多糖对2型糖尿病大鼠肾脏的保护作用陆小元(江苏省姜堰市中医院药剂科,江苏姜堰,225400)摘要:目的观察麦冬多糖对2型糖尿病大鼠肾脏的保护作用。方法采用链脲佐菌素(STZ)诱导2型糖尿病大鼠模型,给予不同剂量麦冬多糖(200、400、800Irlg/kg)治疗6周后,检测各组大鼠空腹血糖(FBG)、肾脏指数、血肌酐(Scr)、尿素氮(BUN)含量,比较各组大鼠肾组织中总抗氧化能力(T-AOC)及过氧化氢(H2O2)的含量,Westernbolt检测肾组织中p47、核因子-KB(NF-KB)蛋白表达。结果模型组大鼠FBG、肾脏指数、Scr、BUN含量明显升高,’肾脏T.AOC明显降低,肾组织p47ph、NF-KB的表达及H202含量显著升高。麦冬多糖能明显降低糖尿病大鼠FBG、肾脏指数、Scr及BUN水平,提高肾脏T.AOC,降低肾脏p47ph、NF—KB的表达及H202的含量。结论麦冬多糖对2型糖尿病大鼠肾脏具有一定的保护作用,其机制可能与其抗氧化、下调糖尿病大鼠肾脏p47ph、NF—KB的表达有关。关键词:麦冬多糖;糖尿病肾病;尼克酰胺腺嘌呤二核苷酸氧化酶;氧化应激中图分类号:R587.1文献标志码:A文章编号:1672—2353(2012)24—0011—04Effectofophiopogon-p0lysaccharideonprotectionofthekidneyintype2diabeticratsLUXiaoyuan(JiangyanHospitalofTraditionalChineseMedicine,Jiangyan,Jiangsu,225400)ABSTRACT:ObjectiveToobserveeffectofophiopogon—polysaccharideonprotectionofkidneyintype2diabeticrats.MethodsStreptozotocin·induceddiabeticratsweregivenophiopogon-polysac-charide(200,400,800mg/kg)for6weeks.Thechangesinbloodglucose(FBG),kidneyweightindex,andserumcreatinine(Scr)andbloodureanitrogen(BUN)weredeterminedaftertreatment.Thetotalantioxidantcapacity(T—AOC)andhydrogenperoxide(H2O2)inrenaltissueweremeasuredandcomparedbetweengroups.Inaddition,theexpressionsofp47。,nuclearfactor—KB(NF-KB)inthekidneytissuesweredeterminedbywesternbolt.ResultsBG,kidneyweightindexandScrandBUNinthemodelgroupincreasedsignificantly,renalT—AOCsignificantlydecreased.ThecontentsofrenalH202andexpressionsofrenalp47,NF—KBincreasedsignificantly.Comparedwithmodelgroup,thelevelofBG,kidneyweightindexandScrandBUNinophiopogon—polysaccharidegroupsdecrea...
