第46卷第1期2023年1月河北农业大学学报JOURNALOFHEBEIAGRICULTURALUNIVERSITYVol.46No.1Jan.2023不同陆地棉品种愈伤组织诱导比较及遗传转化验证鲍红帅,尚红燕,王国宁,王省芬,马峙英,吴金华(河北农业大学农学院/华北作物改良与调控国家重点实验室/河北省作物种质资源重点实验室,河北保定071000)摘要:愈伤组织的诱导能力受基因型的影响,基因型的拓宽可促进棉花遗传转化的发展。本文以8个陆地棉品种的下胚轴为材料,研究不同品种愈伤组织的增殖和状态差异,然后以可再生的品种为受体进行GhAPX基因的遗传转化。结果表明:8个品种均能诱导出愈伤组织,且2号和3号陆地棉品种愈伤组织增殖速度快,依次为7号、1号和8号,除5号和6号外,其它品种均获得再生植株;获得以8号品种为受体转GhAPX基因且经PCR鉴定的阳性苗3株,Real-timePCR分析显示GhAPX基因在3株阳性植株中都上调表达。本研究为棉花遗传转化中受体基因型的拓宽奠定了基础。关键词:陆地棉;下胚轴;愈伤组织;基因型中图分类号:S188开放科学(资源服务)标识码(OSID):文献标志码:AComparisonofcallusinductionabilityofdifferentuplandcottonvarietiesandverificationofgenetictransformationBAOHongshuai,SHANGHongyan,WANGGuoning,WANGXingfen,MAZhiying,WUJinhua(CollegeofAgronomy/StateKeyLaboratoryofNorthChinaCropImprovementandRegulation/KeyLaboratoryforCropGermplasmResourcesofHebei,HebeiAgriculturalUniversity,Baoding071000,China)Abstract:Theinductionabilityofcallusisaffectedbygenotype.Andbroadeningofgenotypeisimportantforthedevelopmentofcottongenetictransformation.Thehypocotylscollectedfromeightuplandcottonvarietieswereusedasmaterialstostudythecallusproliferationandcallusstatus.ThenGhAPXgenewastransformedintotheregeneratedvariety.Theresultsshowedthatcalluscouldbeinducedfromthehypocotylsofalltheeightvarieties,andthecallusofvarietyNo.2andNo.3proliferatedrapidly,followedbyvarietyNo.7,No.1andNo.8.RegeneratedplantswereobtainedexceptvarietyNo.5andNo.6.Finally,weobtainedthreepositiveplantsofvarietyNo.8whichwereidentifiedbyPCRofthetransformedGhAPXgene.Real-timePCRanalysisfurtherconfirmedhighertranscriptsofGhAPXgeneinallthethreepositiveplants.Thisstudylaidafoundationforbroadeningreceptorgenotype...
