运动以Perilipins和FATP为靶点对脂滴代谢与UPR的调控作用PerilipinsandFATPServeasTargetsofExercisetoRegulateLipidDropletMetabolismandUPR金海秀1,2,3,漆正堂1,2,MOLLEREAUBertrand3,丁树哲1,2*JINHaixiu1,2,3,QIZhengtang1,2,MOLLEREAUBertrand3,DINGShuzhe1,2*摘要:肥胖常常伴随脂滴在肝脏、脂肪和骨骼肌等器官或组织的过度堆积和异位堆积,是引起2型糖尿病的主要危险因素。在脂滴代谢过程中,围脂滴蛋白家族(perilipins)和脂肪酸转运蛋白(fattyacidtransportationprotein,FATP)发挥关键作用,可以影响脂滴、内质网和线粒体等细胞器的功能,进而调节细胞代谢。已有研究表明,高脂膳食和2型糖尿病中伴随着由脂滴代谢引起的内质网应激和非折叠蛋白反应(unfoldedproteinresponse,UPR),而perilipins和FATP在其中起到主导作用。运动是减肥和治疗2型糖尿病的有效手段之一,不同的运动方式可以激活内质网应激反应,通过调节perilipins和FATP蛋白家族的表达情况介导脂滴代谢,影响骨骼肌的功能。因此,研究不同的运动方式对骨骼肌细胞脂滴代谢的影响情况,有利于分析运动对骨骼肌的调节作用。关键词:脂滴代谢;围脂滴蛋白家族;脂肪酸转运蛋白;非折叠蛋白反应;运动;肥胖和2型糖尿病Abstract:Obesityischaracterizedbytheexcessiveandectopicaccumulationoflipiddroplets(LD)inliver,fattissueandskeletalmuscle.Itisamainriskfactorfortype2diabetes.Perilipinsproteinfamilyandfattyacidtransportationproteins(FATP)playkeyrolesinthelipiddropletmetabolism.TheycanaffectthefunctionsofLDs,endoplasmicreticulum(ER),mitochondriaandotherorganelles,therebyregulatingthecellmetabolism.RecentresearcheshaveshownthattheperilipinsandFATP-mediateddysfunctionofLDmetabolismcantriggerERstressandacti‐vatetheunfoldedproteinresponse(UPR).AsakeydownstreammoleculeintheIRE1/RIDDpathway,FATPcloselylinksLDmetabolismwithERstress.Exerciseisanefficientwaytoloseweightandtreattype2diabetes.Differentexercisecannotonlyaffecttheexpressionofperili‐pinsandFATPproteinsinskeletalmuscle,butalsoactivateERstress,therebyregulatingLDmetabolismandaffectingthefunctionofskeletalmuscle.Therefore,studyingtheeffectsofdif‐ferentexercisemethodsonlipidmetabolismisveryhelpfultoanalyzetheregulatorye...
