·基础研究·*基金项目:云南省科技厅课题(202102AA100021-11)**通信作者:孔洁,E-mail:peanyt3402@21cn.comBMP9修饰对BMSCs修复颌骨缺损的作用及机制*阮吉,孔洁**,杨贞娴,吴京蔓,赵悦涛,刘春海(红河州第一人民医院云南红河661000)[摘要]目的:探究BMP9修饰对BMSCs修复颌骨缺损的作用机制。方法:分离、培养并鉴定BMSCs,检测BMSCs增殖能力,茜素红染色和油红O染色检测BMSCs成骨、成脂能力。RT-PCR检测BMSCs中BMP9表达。将大鼠随机分为Control组、BMSCs+Ad-GFP组、BMSCs+Ad-BMP9组,每组10只。采用HE染色检测骨缺损修复程度,比较新生骨面积百分比。蛋白质免疫印迹法检测组织中ALP、OCN蛋白表达。结果:BMSCs表面CD90、CD105表达升高,CD45、CD34表达降低,证明BMSCs保持了间充质干细胞的活性。BMSCs在0~7d时增殖能力明显变强,7~9d时BMSCs生长达到平台期,且BMSCs分泌生成的骨结节及脂滴被染成红色。RT-PCR结果显示BMSCs+Ad-BMP9组中BMP9mRNA表达较BMSCs+Ad-GFP组明显增多(P<0.05)。与Control组相比,BMSCs+Ad-GFP组和BMSCs+Ad-BMP9组大鼠牙槽骨组织学评分、牙槽骨缺损区新生骨面积及成骨相关基因ALP、OCN蛋白表达均明显增加(P<0.05),且BMSCs+Ad-BMP9组均明显高于BMSCs+Ad-GFP组(P<0.05)。结论:BMSCs可修复颌骨缺损,BMP9修饰BMSCs可促进BMSCs对颌骨缺损的修复作用。[关键词]骨形态发生蛋白9(BMP9)修饰;骨髓间充质干细胞(BMSCs);颌骨缺损;成骨能力;成脂能力[中图分类号]Q813.1[文献标识码]Adoi:10.3969/j.issn.1003-1634.2023.06.002EffectandmechanismofBMP9modificationonrepairingjawdefectswithBMSCs.RUANJi,KONGJie**,YANGZhen-xian,WUJing-man,ZHAOYue-tao,LIUChun-hai.TheFirstPeople’sHospitalofHonghePrefecture,YunnanHonghe661000,China.[Abstract]Objective:ToexplorethemechanismofBMP9modificationinrepairingjawdefectswithBMSCs.Meth-ods:BMSCswereisolated,culturedandidentified,andtheirproliferativeabilitywasdetected.AlizarinredstainingandoilredOstainingwereusedtodetecttheosteogenicandlipogenicabilityofBMSCs.RT-PCRwasperformedtodetectBMP9expres-sioninBMSCs.TheratswererandomlydividedintoControlgroup,BMSCs+Ad-GFPgroupandBMSCs+Ad-BMP9group,10ratsineachgroup.TheHEstainingmethodwasusedtodetecttheextentofbonedefectrepairbycomparingthepercentageo...
