4期ArylHydrocarbonReceptor-mediatedCardiacToxicityinAdultZebrafishInducedbyTetrabromobisphenolASHANJing1,2,MAXiaofa1,2,WUMengyu1,2,LINYujia1,2,WANGYi1,2,WANGRui1,2,XUHaiming1,2(1.SchoolofPublicHealthandManagement,NingxiaMedicalUniversity,Yinchuan750004,China;2.TheKeyLaboratoryofEnvironmentalFactorsandChronicDiseaseControlofNingxia,Yinchuan750004,China)Abstract:ObjectiveInthisstudy,zebrafish(Daniorerio),amodelorganism,wasusedasanexperimentalanimal,andCH223191(CH),anaromatichydrocarbonreceptor(AhR)inhibitor,wasusedtoexploretheroleofAhRinthecardiactoxiceffectoftetrabromobisphenolA(TBBPA)onzebrafish.MethodsAdultzebrafishwererandomlydividedintosixgroupsforexposure,namelysolventcontrolgroup(DMSO),CHgroup(0.05μmol·L-1),TBBPAexposuregroups(10,100,1000nmol·L-1),andCH+TBBPA(1000nmol·L-1)group.After30daysofexposure,thebodyweightandbodylengthweremeasured.TheexpressionofAhR-relatedgenes(ahr2,cyp1a1andcyp1a2),cardiacfunction-relatedgenes(sox9b,nkx2.5,gate4,myh6,tbx5andvmhc)andreactiveoxygenspecies-relatedgenes(nrf2,gpx1a,sod1,sod2andcat)incardiactissueweredetectedbyqRT-PCR.Theantioxidantenzymes(SOD,CATandGSH-Px/GPX)activitiesinlivertissueweretestedbycommercialkits.ResultsComparedwiththeDMSOcontrolgroup,therewasnosignificantdifferenceinbodyweightandbodylengthinallexposuregroups(Pall>0.05).TheresultsofqRT-PCRshowedthatAhR-relatedgenes(ahr2,cyp1a1andcyp1a2)wasdown-regulatedintheCHgroup,whileup-regulatedin10,100,1000nmol·L-1TBBPAgroups(Pall<0.05).Comparedwiththe1000nmol·L-1TBBPAexposuregroup,theseAhRrelatedgenesweredecreasedtoacertainextentintheCH+1000nmol·L-1TBBPAgroup(Pall<0.05).Thecardiacfunction-relatedgenes(sox9b,nkx2.5,gate4,myh6,tbx5andvmhc)weredown-regulatedinthe1000nmol·L-1TBBPAgroup(Pall<0.05).Thereactiveoxygenspecies-relatedgenes(gpx1a,sod1,sod2andcat)weredown-regulatedwhilenrf2wasup-regulatedinthe1000nmol·L-1TBBPAgroup(Pall<0.05).Theresultsoftheantioxidantenzymeact...
