试验研究NO.02202389饲料研究FEEDRESEARCH1株耐酸秸秆纤维素降解菌株的筛选、鉴定及其内切葡聚糖酶的异源表达研究张超1,2李晓意1杨吉1武晓乐1张洋3李孟欣1刘宇欣1李凯悦1(1.衡水学院生命科学学院,河北衡水053000;2.河北省果蔬发酵技术创新中心,河北衡水053000;3.河北农业大学生命科学学院,河北保定071001)摘要:试验旨在解决农作物秸秆饲料化过程中,秸秆纤维素难降解、适口性差和营养价值低等问题。试验以玉米秸秆粉为唯一碳源,从堆砌秸秆的土壤中分离筛选出1株耐酸且具有高纤维素酶活性的菌株HSU-20SJ。经3,5-二硝基水杨酸法测定纤维素酶活性,利用形态学、生理生化和16SrDNA进行种属鉴定,通过分子克隆技术,构建内切葡聚糖酶的大肠杆菌异源表达系统。结果显示,菌株HSU-20SJ粗酶液酶活为3.23U/mL,最适反应pH值为5.0。该菌株被鉴定为贝莱斯芽孢杆菌(Bacillusvelezensis),从菌株HSU-20SJ基因组中获得内切葡聚糖酶基因eg20SJ,异源表达后获得重组酶,分子量约为55kDa,粗酶液酶活为6.28U/mL。研究表明,试验在获得耐酸、高纤维素酶活菌株基础上,实现了内切葡聚糖酶基因的异源表达,可为纤维素酶的规模化生产和应用提供参考。关键词:秸秆;纤维素;降解;内切葡聚糖酶;异源表达中图分类号:S816.7文献标识码:A文章编号:1002-2813(2023)02-0089-05Doi:10.13557/j.cnki.issn1002-2813.2023.02.018Studyonscreeningandidentificationofanacid-tolerantstrawcellulose-degradingbacteriaandheterologousexpressionofendoglucanaseZHANGChaoLIXiao-yiYANGJiWUXiao-leZHANGYangLIMeng-xinLIUYu-xinLIKai-yueAbstract:Thestudywastosolvetheproblemsofstrawcellulosedifficulttodegrade,poorpalatabilityandlownutritionalvalueinprocessofstrawfeed.Usedcornstalkpowderassolecarbonsource,astrainHSU-20SJwithhighcellulaseactivityandacid-tolerancewasisolatedfromsoilpiledwithstraw.Cellulaseactivitywasdeterminedby3,5-dinitrosalicylicacidmethod,speciesidentificationwascarriedoutbymorphology,physiologyandbiochemistryand16SrDNA,andtheheterologousexpressionsystemofendoglucanaseinEscherichiacoliwasconstructedbymolecularcloningtechnology.TheresultsshowedthatthecrudeenzymeactivityofstrainHSU-20SJwas3.23U/mL,andtheoptimumreactionpHwas5.0.ThestrainwasidentifiedasBacillusvelezensis.Theendoglucanas...
